Evaluation of the priming methods associated with refilling in the FRP at mammalian glutamatergic synapses has not been trivial because releasecompetent SVs are heterogeneous in release probability and their recovery kinetics (26, 27). The present study indicates that such SVs are fully matured only after they are positioned close to the Ca2 supply. We demonstrate that the time course for such complete maturation or superpriming of newcomer SVs is slower ( = three.6 s) than that of cytoskeletondependent conversion of reluctant SVs into FRP SVs ( = 60 ms) (6). For that reason, we propose a twostep model for refilling from the FRP: rapid “positional priming,” which brings vesicles closer to Ca2 sources, followed by slower superpriming, which enhances the Ca2 sensitivity of vesicles.
